Co-Kulturen) wurde eine mikrofluidische Sonde eingesetzt. Um den mikroskalierten lokalen Einschluss der Biochemikalien zu realisieren, wurde ein gleichzeitiger Injektions- und Aspirationsprozess angewandt- hydrodynamic flow confinement. Der Liquid-Handling-Prozess wurde mit 4 neMESYS Niederdruck-Spritzenpumpen realisiert. Gro\u00dfe Lysatvolumina (&gt;100 \u00b5l) wurden mit einer Spritze durch die assoziierende Aspirationspumpe gesammelt. Kleinere Probenvolumina (&lt;10 \u00b5L) wurden direkt am mikrofluidischen Sondenkopf mittels neMESYS Niederdruck-Spritzenpumpe gesammelt und anschlie\u00dfend in ein PCR-Gef\u00e4\u00df injiziert. Dazwischenliegende Lysatvolumina (\u2248 50 \u00b5L) wurden direkt in die Probenschleife des Qmix V extrahiert.<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter size-full wp-image-4592\" src=\"https:\/\/cetoni.de\/wp-content\/uploads\/blog_local_lysis.png\" alt=\"\" width=\"1280\" height=\"835\" srcset=\"https:\/\/cetoni.com\/wp-content\/uploads\/blog_local_lysis.png 1280w, https:\/\/cetoni.com\/wp-content\/uploads\/blog_local_lysis-300x196.png 300w, https:\/\/cetoni.com\/wp-content\/uploads\/blog_local_lysis-1024x668.png 1024w, https:\/\/cetoni.com\/wp-content\/uploads\/blog_local_lysis-768x501.png 768w\" sizes=\"auto, (max-width: 1280px) 100vw, 1280px\" \/><\/p>\n<p>Zusammenfassend l\u00e4sst sich sagen, dass basierend auf dem neMESYS Spritzenpumpensystem und den Features der Automatisierungssoftware QmixElements, ein neuartiges Setup f\u00fcr die Zelllyse und Probensammlung realisiert werden konnte. Dabei unterst\u00fctzen 5 Spritzenpumpen die notwendige Injektion und Absaugung der unabh\u00e4ngigen Str\u00f6me verschiedener Reagenzien, die eine kontinuierliche Zelllyse zur Generierung hochkonzentrierter Lysate f\u00fcr die DNA Analyse gew\u00e4hrleisten.
